Rope is definitely based on them! It's actually based on a play where the boys are more implied to be explicitly homo in a slightly more sinister way, as they're also stated as making passes at their poor manservant. There are double entendres and an implied crush and perhaps relationship with a former professor(Who in this version is a dreamboat, WWI veteran with a bum leg), which adds another layer to the whole 'gay as evil' thing, given the fact that the boys were sexually abused irl. The author of this play was an irl homo, for context.
My understanding is that quite a few adaptations went in with the idea of 'disturbingly close friendship 4 creepy' instead of homo.
Hitchcock found the homo so important to the plot and characters that he a) Didn't play it up for sensationalised creeps, and b) hired explicitly gay actors so that the chemistry would be authentic. Keep in mind that the Hays Code forbade showing homo at all. In a subtle chamber drama like that, I'd say it made all the difference.

I thought Rope was the best Hitchcock I've seen, liked it more than Vertigo and Psycho. When I originally decided to watch it I chose to do so because I'd heard it was shot like one long take, and I thought the closed room nature of the film played to that strength. I also liked how organic the film felt, there are a few scenes where the two leads are having a conversation of their own while the party guests are talking and it almost muffles them. Really adds to the feeling of the movie events happening in real time.

Hitchcock knew how to use gay subtext for predatory and creep reasons, and did it well. Predatory homos can be hot as well, so not knocking that, but I found it really sweet that he saw the potential for drama in the gay couple in Rope, without having to explicitly play it up for creep factor. It's one of the more 'low-key' adaptations I've seen, I think it works very well because the characters seem like people one would know irl.
At one point, I recall it's made clear that there's _one_ bed in the apartment. One. A single one. Now why would that be :D
